The study was carried out to investigate the effects of Sophora subporsrate polysaccharide on the apoptosis of thymocyte and spleen lymphocytes in mouse induced by dexamethasone,and it was investigated and detected by methods of agarose gel electrophoresis,transmission electron microscopy,flow cytometry,apoptosis mode of hymocyte and spleen lymphocytes was constructed by intraperitoneal injection on mouse.DNA agarose electrophoresis showed that the apoptosis control group and Sophora subporsrate polysaccharide groups with different doses emerged a typical ladder.The characteristics of nucleus chromatin of thymocyte and spleen lymphocytes showed the shape of a half moon,'C' and apoptotic bodies and peritoneal macrophage phagocytizing apoptotic bodies has been found under transmission electron microscopy.Flow cytometry analysis showed that apoptosis rate in group of Sophora subporsrate polysaccharide group with different doses decreased significantly compared with the apoptosis control group(P0.01),and with the increasing dose of Sophora subporsrate polysaccharide,the apoptosis ratio was decreased.The lowest rate of apoptosis was observed at the dose of 400 mg/kg Sophora subporsrate polysaccharide.In conclusion,Sophora subporsrate polysaccharide at 50-400 mg/kg dose had significant inhibitory effects on the apoptosis of thymocyte and spleen lymphocytes in mouse induced by dexamethasone.
