Sirtuins are regulators of fundamental biological processes and the role of sirtuins in longevity and age-related diseases has been investigated. The longevity effect of sirtuins has been investigated in many species including yeast, worms, flies, bees, fish, and mice, although their longevity effects have been the subject of some controversies. In addition, sirtuin activators including calorie-restricted conditions, small molecules, natural compounds, and components related to NAD+ salvage pathways have been also investigated in relation to their capacity to affect life span extension. In this chapter, we reviewed the longevity effect of sirtuins and their activators.
