Objectives To study the effect of Bacteroides thetaiotaomicron(BT) intervention on the blood glucose,body weight and C-peptide level in diabetic mice.Methods(1) Diabetic mouse models were injected alloxan for 15 days(200 mg / kg intraperitoneal injection) and divided into four groups: control group(n = 10),bacteria-injected control group(n = 10),alloxan-induced diabetic model group(n = 11) and bacteria-injected model group(n = 11).(2) ATCC 29148 standard strains were cultured on BHI solid medium;bacteria count was determined using nephelometry method.(3) The bacteria-injected control and model groups were injected Bacteroides thetaiotaomicron suspension,and the changes of body weight,fasting blood glucose(FBG) levels of four groups were detected.The amount of bacteroides was determined by Real-time PCR;enzyme-linked immunosorbent assay(ELISA) was used to detect the serum Cpeptide levels.Results(1) Comparing with the control group,BT colonization appeared at Day 3 and still maintained at Day 15 in bacteria-injected control group.Comparing with the model group,BT colonization appeared at Day 15 in alloxan-induced diabetic model group.(2) Linear regression correlation analysis showed that Bacteroides thetaiotaomicron was negatively correlated with body weight(r =-0.70,P 0.05) while positively correlated with FBG(r = 0.71,P 0.05);C-peptide showed a negative correlation(r =-0.62,P 0.05);HOMA-IR was positively correlated(r = 0.55,P 0.05) and HOMA-IS showed a negative correlation(r =-0.43,P 0.05).Conclusion(1) Bacteroides thetaiotaomicron can colonize the intestine via exogenous administration.(2) Bacteroides thetaiotaomicron number changes are associated with diabetes.
