Objective To establish a adriamycin(ADR)-induced myocarditis model in primary culture neonatal Sprague Dawley rat cardiac myocytes and provide basis for mechanism research and drug therapy of ADR-induced cardiotoxic myocarditis.Meshods 10 ng·ml(-1)~100 μg·ml(-1) ADR were injected in injured primary culture neonatal SD rat cardiac myocytes,and with the effect was observed with different concentrations or different time(24,48 or 72 h),the apoptotic cardiac myocyte was shown by using MTT assay.Lactate dehydrogenase(LDH) and glutamic oxaloacetic transaminase(GOT) of superior fluid were measured. Results ADR could increase cell apoptosis,at the same time,but the survival rate of cardiac myocytes decreased significantly when the concentration was above 10 ng/ml.And the inhibition action was concentration-dependent.ADR could promote the release of LDH and GOT of superior fluid in the concentration-dependent manner.Meantime,the longer ADR contacted with cardiac myocytes,the higher the survival rate was,and the lower LDH and GOT were.Conclusion ADR can damage cardiac myocytes of primary culture,and the damage is in direct proportion to the concentration of ADE and acting time.
