Pharmacokinetics of piperine capsules in healthy volunteers

Objective To establish an HPLC-MS/MS method for the determination of piperine in human plasma and to study its pharmacokinetics in healthy volunteers.Methods Pharmacokinetics of piperine were studied in 8healthy vol-unteers,following a single oral dose and multiple doses(20mg).The drug concentrations in the serum samples were assayed by HPLC-MS/MS.The pharmacokinetic parameters were calculated by DAS 2.1.1soft ware.Results Ser-um concentrations ranging from 0.50to 800.00ng.mL-1 were found to have good linearity.The precisions of within-day and the recovery rate were all coincided with the clinical pharmacokinetics.The pharmacokinetics of piperine fit in a two-compartment open model,the mean peak concentrations(Cmax)in the serum was(290.00±42.47),(595.39± 108.61)ng.mL-1;the area under the concentration-time curve(AUC)was(5 932.26±108 2.01),(15 796.67± 5 050.39)ng.h.mL-1;and the elimination half-life(t1/2β)was(13.26±1.91)and(15.82±4.95)h,respectively.Conclusion The pharmacokinetics of piperine fit in a two-compartment open model and there is no accumulation after a multiple oral dose of piperine in 7days.
