Abstract Extensive training and significant resources are currently required for oligosaccharide synthesis, causing a bottleneck in glycosciences. Many, if not all, current synthetic methods still rely upon specialized knowledge of carbohydrate chemistry. This limits availability of synthetic glycans and inhibits their industrial production and application. Traditional manual synthesis has expanded in recent years giving scientists more accessibility to creating both simple and relatively complex oligosaccharides. The application of potentially transformative automated technologies is expected to expand the access to synthesizing oligosaccharides and carbohydrates in general. Described herein are advances that have been made in the development of one such technology, HPLC-assisted oligosaccharide synthesis or HPLC-A, since its invention in 2012.
