This paper adopted colorimetry and spectrofluorimetry to analyze the effect of high-intensity pulsed electric field on the activity and conformation of horseradish peroxidase (HRP). The results showed that the activity of HRP decreased as electric field intensity and pulse number increased. When the electric field intensity was 20 kV/cm, the activity of HRP decreased by 15% and 55% at 20 and 80 pulse numbers, respectively. And the fluorescence of HRP quenched in different degrees by pulsed electric field. The place of peak fluorescence intensity changed on high electric field intensity. Besides, the trend of inactivation and fluorescence quenching was similar, however, the relation between them is nonlinear. The same critical electric field intensity (about 10 kV/cm) existed in the processes of inactivation and fluorescence quenching .All these indicated that changes of HRP conformation were on intimate terms with the inactivation.
