Quantitative determination of chlorogenic acid,ferulic acid and glycyrrhizic acid in Si-Miao-Yong-An decoction by capillary zone electrophoresis

Objective:To develop a capillary zone electrophoresis method for the simultaneous quantitative determi- nation of chlorogenic acid,ferulic acid and glycyrrhizic acid in Si-Miao-Yong-An decoction.Method:A uncoat- ed fused silica capillary(57 cm×50μm,valid length 50 cm)was used to separate the components;Para-amino- benzoic acid was used as internal standard;the running buffer contained 50 mmol·L~(-1)borax with 12.5 mmol·L~(-1) tri-hydroxyl-methyl-aminomethane(Tris,pH 9.95)under the voltage of 22 kV at 25℃;Injection time was 5 s with 138 kPa Nitrogen pressure,Diode assay detector was used to record the spectrum and to monitor at 254 nm;The running time lasted for 20 minutes.Results:The linear calibration range was 0.09-1.50 mg·mL~(-1)for chlorogenic acid,0.01-0.16 mg·mL~(-1)for ferulic acid and 0.04-0.66 mg·mL~(-1)for glycyrrhizic acid;the correlation coef- ficient was 0.9994 for chlorogenic acid,0.9994 for ferulic acid and 0.9993 for glycyrrhizic acid;the average recov- ery was 97.8% for chlorogenic acid,98,l%for ferulic acid and 96.3% for glycyrrhizic acid.Conclusion:The ana- lytical results demonstrated the method is simple,rapid and well reproducible.
