摘要
A general protocol was developed for the synthesis of 3-sulfonyl-2-aminopyridines, valuable building blocks in drug development. The three-component one-pot coupling between various N,N-dimethylacroleines, sulfonylacetonitriles, and primary amines/ammonia yields the desired products that are isolated with high purity and excellent yields without the need of column chromatography. The scope of the reaction demonstrates remarkable versatility and scalability, making it highly suitable for large-scale applications in process and medicinal chemistry.
